The assay uses the sandwich enzyme immunoassay technique on a pre-coated 96-well microplate, employing a biotinylated anti-ATP1B3 antibody and an HRP-avidin conjugate. A TMB substrate produces a blue colour that turns yellow upon addition of the stop solution, and absorbance is read at 450 nm using a colorimetric microplate reader.

Sensitivity is reported as <0.066 ng/ml with an assay range of 0.156-10 ng/ml, and total assay time is 4 hours 30 minutes. Manufacturer-reported average recoveries are 91% for serum, 90% for EDTA plasma and 90% for heparin plasma.
