Mouse Aryl Hydrocarbon Receptor Nuclear Translocator ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com enables the in vitro quantitative determination of mouse HIF-1 beta in tissue homogenates or other biological fluids. The assay uses a 96-well microtiter plate pre-coated with an antibody specific for HIF-1 beta. Standards and test samples are pipetted into the wells, where HIF-1 beta present is captured by the immobilized antibody. Following incubation and washing, a Biotinylated Anti-HIF-1 beta Antibody is added, then an HRP-Avidin conjugate. TMB substrate solution is used to visualise the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution. The intensity of yellow is proportional to the amount of HIF-1 beta captured in each well, and concentration is calculated by reading the O.D. absorbance at 450 nm using a colorimetric microplate reader and referring to the standard curve.

The kit demonstrates a sensitivity of <0.054 ng/ml and a working range of 0.156–10 ng/ml, with a total assay time of 4 hours 30 minutes on a pre-coated microplate platform of 12 x 8 well strips. Reported average recovery values are 91% for serum, 90% for EDTA plasma, and 84% for heparin plasma. Kit components should be stored at +4°C or -20°C as instructed, and the kit must not be used past its expiration date.
