Mouse Arachidonate-5-Lipoxygenase ELISA Kit

From Antibodies.com, this sandwich ELISA kit enables the in vitro quantitative determination of mouse 5 Lipoxygenase/5-LO (Arachidonate 5-Lipoxygenase) in tissue homogenates, cell lysates, or other biological fluids. The assay uses a pre-coated 96-well microplate arranged as 12 x 8 well strips, with quantification performed by reading the O.D. absorbance at 450 nm in a colorimetric microplate reader.

The procedure employs the sandwich enzyme immunoassay technique, in which a capture antibody specific for 5-LO is immobilised on the plate. Standards and samples are added and any 5-LO present is bound by the immobilised antibody. After washing, a biotinylated anti-5-LO antibody is added, followed by an HRP-Avidin conjugate. TMB substrate is then introduced; HRP catalysis yields a blue product which turns yellow upon addition of the acidic stop solution. The intensity of yellow is proportional to the amount of 5-LO captured in each well, and the concentration is calculated by reference to the standard curve.

The kit has a reported product range of 0.156–10 ng/ml and a sensitivity of 0.058 ng/ml. The complete assay takes 4h 30m and is suitable for cell lysates and tissue homogenates.
