Mouse Aquaporin 3, Gill Blood Group ELISA Kit

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of mouse Aquaporin 3 (Gill Blood Group; also known by the synonyms AQP-3, AQP3, Aquaglyceroporin-3 and Aquaporin-3). The kit is suitable for serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids.

An antibody specific for Aquaporin 3 is pre-coated onto a microplate arranged as 12 x 8 well strips. Standards and test samples are added, captured Aquaporin 3 is detected with a Biotinylated Anti-Aquaporin 3 Antibody followed by an HRP-Avidin conjugate, and the reaction is visualised with TMB substrate, producing a blue product that changes to yellow after addition of acidic stop solution. Absorbance is read at 450 nm in a colorimetric microplate reader, and Aquaporin 3 concentration is calculated from the standard curve. Reported assay time is 4h 30m. The measurement range is 0.156-10 ng/ml, with a sensitivity of <0.058 ng/ml.

Spike-and-recovery testing (n=5) gave averages of 91% for serum (range 80-102%), 90% for EDTA plasma (range 81-100%) and 84% for heparin plasma (range 80-89%). Linearity was demonstrated across 1:2 to 1:16 dilutions for serum, EDTA plasma and heparin plasma samples. Supplied components include the pre-coated microplate strips, lyophilized standard, Detection Solutions A and B, 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution and plate sealers, to be stored at +4°C or -20°C as instructed.
