Mouse Apoptotic Peptidase Activating Factor 1 ELISA Kit

Antibodies.com offers a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) for the in vitro quantitative determination of mouse Apoptotic Peptidase Activating Factor 1 (APAF1) in tissue homogenates, cell lysates and other biological fluids. The kit is reactive with mouse samples and utilises a pre-coated 96-well microplate arranged as 12 x 8 well strips.

An antibody specific for APAF1 is pre-coated onto the plate; standards and samples are added and bound by the immobilised antibody. Wells are washed and incubated with Biotinylated Anti-APAF1 Antibody, followed by HRP-Avidin conjugate. TMB substrate is used to visualise the enzymatic reaction, producing a blue product that turns yellow upon addition of acidic stop solution. Absorbance is read at 450 nm on a colourimetric microplate reader. The total assay time is 4h 30m, and the assay range is 78.1-5000 pg/ml with a sensitivity of <28 pg/ml.

Reported average recovery values for serum (n=5) are 91%, EDTA plasma (n=5) 90%, and heparin plasma (n=5) 84%, with linearity data provided across dilution factors of 1:2, 1:4, 1:8 and 1:16 for serum, EDTA plasma and heparin plasma samples. Kit components include a pre-coated microplate, lyophilised standard, detection solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers, to be stored at +4°C or -20°C as instructed.
