Manufactured by Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of mouse APE1 in tissue homogenates, cell lysates or other biological fluids. The assay employs a pre-coated 96-well microplate onto which an antibody specific for APE1 has been immobilised. Standards and test samples are added to the wells, and APE1 present in each sample is bound by the immobilised antibody.

After incubation and washing, a Biotinylated Anti-APE1 Antibody is added, followed by an HRP-Avidin conjugate. A TMB substrate solution is used to visualise the enzymatic reaction, producing a blue-coloured product that changes to yellow after addition of acidic stop solution. The density of yellow is proportional to the amount of APE1 captured in each well, and the concentration of APE1 is calculated by reading the O.D. absorbance at 450 nm in a microplate reader and referring to the standard curve.

Reported sensitivity is <0.051 ng/ml with a quantitative range of 0.156–10 ng/ml and a total assay time of 4h 30m. The kit is intended for use with a colorimetric microplate reader, and components are stored at +4°C or −20°C as instructed. Reported average spike-and-recovery values were 91% for serum (80%–102%, n=5), 90% for EDTA plasma (81%–100%, n=5), and 84% for heparin plasma (80%–89%, n=5).
