From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of mouse Pancreatic alpha Amylase in serum, plasma, tissue homogenates and other biological fluids. The assay employs the sandwich enzyme immunoassay technique, with an antibody specific for Pancreatic alpha Amylase pre-coated onto a 96-well microtiter plate. After binding and washing, a Biotinylated Anti-Pancreatic alpha Amylase Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate is then used to visualise the enzymatic reaction, yielding a blue product that changes to yellow upon addition of acidic stop solution. The intensity of yellow is proportional to the amount of target captured in each well, and concentration is calculated from the O.D. absorbance at 450nm against a standard curve.

Reported performance characteristics include a sensitivity of <4.1 pg/ml and a working range of 9.38-600 pg/ml. The kit is intended for use with mouse samples and has an assay time of 4h 30m. Recovery values are reported as 80%-102% in serum (n=5, average 91%), 81%-100% in EDTA plasma (n=5, average 90%) and 80%-89% in heparin plasma (n=5, average 84%). Linear dilution behaviour has been characterised for serum, EDTA plasma and heparin plasma at 1:2, 1:4, 1:8 and 1:16 dilutions. The kit is supplied with a pre-coated microplate, lyophilised standard, detection solutions, wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers, and is intended for research use only.
