Description

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of mouse alpha 2 Macroglobulin in serum, plasma, urine or other biological fluids. The kit targets Alpha 2 Macroglobulin in mouse samples and is run on a pre-coated 96-well microplate (12 x 8 well strips), with absorbance read on a colorimetric microplate reader at 450 nm.

The assay employs the sandwich enzyme immunoassay technique. An antibody specific for alpha 2 Macroglobulin is pre-coated onto the microtiter plate; standards and test samples are added and the target present in each sample is bound to the wells by the immobilized antibody. Following incubation and washing, a Biotinylated Anti-alpha 2 Macroglobulin Antibody is added, which binds the captured target in each well. An HRP-Avidin conjugate is then added and the plate incubated. After further washing, TMB substrate solution is used to visualise the HRP enzymatic reaction by catalysis, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of alpha 2 Macroglobulin captured in each well, and the concentration is calculated by reference to the standard curve.

Sensitivity is reported as <3.7 ng/ml, with an assay range of 9.38-600 ng/ml and a total assay time of 4h 30m. In undiluted samples, average recovery is 91% in serum (range 83-100%), 96% in EDTA plasma (range 90-103%) and 94% in heparin plasma (range 90-99%). Linearity has been demonstrated across serial dilutions from 1:2 to 1:16 in serum, EDTA plasma and heparin plasma. Kit components are to be stored at +4°C or -20°C as instructed and the kit should not be used past its expiration date.
