Antibodies.com supplies this Mouse Alpha 1 Antitrypsin ELISA Kit for the in vitro quantitative determination of alpha 1 Antitrypsin in mouse serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The assay uses a sandwich ELISA format on a pre-coated 96-well microplate arranged as 12 x 8 well strips, with absorbance read in a microplate reader at 450 nm.

The standard curve supports a quantitative range of 3.12–200 ng/ml with a reported sensitivity of <1.42 ng/ml. Total assay time is approximately 4 hours 30 minutes. In the workflow, standards and samples are added to the pre-coated wells; following incubation and washing, a biotinylated anti-alpha 1 Antitrypsin detection antibody is added, followed by an HRP-avidin conjugate. TMB substrate produces a blue product that turns yellow after the acidic stop solution, with the intensity of the yellow colour proportional to the alpha 1 Antitrypsin captured in each well.

Spike-and-recovery performance has been characterised (n=5), with average values of 91% in serum, 90% in EDTA plasma and 84% in heparin plasma. Linearity has been examined across 1:2, 1:4, 1:8 and 1:16 dilutions in serum, EDTA plasma and heparin plasma (n=5). Kit components are intended for storage at +4°C or -20°C as instructed in the protocol manual, and the kit must not be used past its expiration date.
