Mouse ADP Ribosyltransferase 1 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com enables the in vitro quantitative determination of mouse ART1 in tissue homogenates or other biological fluids. The assay employs a pre-coated 96-well microplate arranged as 12 x 8 well strips, with an antibody specific for ART1 immobilised on the wells. Standards and test samples are added; ART1 present in each sample is captured by the immobilised antibody. Following incubation and washing, a Biotinylated Anti-ART1 Antibody is added, which binds the captured ART1. HRP-Avidin conjugate is then added, and after further incubation and washing, TMB substrate solution visualises the HRP enzymatic reaction, producing a blue product that changes to yellow upon addition of acidic stop solution. The intensity of yellow is proportional to the amount of ART1 captured, and the concentration is calculated by reading O.D. absorbance at 450 nm in a colorimetric microplate reader and referring to the standard curve.

The quantitative sandwich assay has an assay range of 0.781–50 ng/ml and a sensitivity of <0.32 ng/ml, with a total assay time of 4 hours 30 minutes. Recovery was characterised in serum (80%–102%, average 91%), EDTA plasma (81%–100%, average 90%) and heparin plasma (80%–89%, average 84%) across tested samples. Kit components are stored at +4°C or −20°C as instructed, and the kit should not be used past its expiration date.
