Mouse Adiponectin ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of mouse Adiponectin in serum, plasma, tissue homogenates, cell lysates, cell culture supernates, and other biological fluids. The assay uses a 96-well microplate pre-coated with an antibody specific for Adiponectin. Standards and samples are added to the wells and Adiponectin is bound by the immobilized antibody. Following incubation and washing, a biotinylated anti-Adiponectin detection antibody is added to form a sandwich complex, after which an HRP-avidin conjugate and TMB substrate are introduced. The enzymatic reaction produces a blue-coloured product that changes to yellow upon addition of acidic stop solution. Absorbance is read at 450 nm using a colorimetric microplate reader and concentration is calculated by reference to a standard curve.

The product range is 31.2-2000 pg/ml with a sensitivity of <14.5 pg/ml. Total assay time is 4 hours 30 minutes. Recovery has been demonstrated in serum (range 80%-102%, average 91%, n=5), EDTA plasma (range 81%-100%, average 90%, n=5), and heparin plasma (range 80%-89%, average 84%, n=5). Linearity across dilutions of 1:2, 1:4, 1:8, and 1:16 was validated in serum (87-91%, 87-107%, 74-101%, 92-97%), EDTA plasma (90-105%, 84-101%, 90-101%, 79-108%), and heparin plasma (84-95%, 92-105%, 82-105%, 89-91%). Kit components are stored at +4°C or -20°C as instructed.
