Mouse Adenylate Cyclase Activating Polypeptide 1, Pituitary ELISA Kit

From Antibodies.com, this sandwich ELISA kit is designed for the in vitro quantitative determination of mouse PACAP-38 in serum, plasma, tissue homogenates and other biological fluids. The assay uses a pre-coated microplate with an antibody specific for PACAP-38 immobilised in the wells. Standards and samples are added, followed by a biotinylated anti-PACAP-38 detection antibody and an HRP-avidin conjugate. A TMB substrate produces a blue colour that turns yellow upon addition of the stop solution; absorbance is read at 450 nm on a colorimetric microplate reader and the PACAP-38 concentration is calculated from the standard curve.

Reported analytical sensitivity is 28 pg/ml with an assay range of 78.1-5000 pg/ml. The protocol takes approximately 4 hours 30 minutes to complete and kit components are stored at +4°C or -20°C as instructed. Average recoveries have been validated at 91% in serum (range 80-102%, n=5), 91% in EDTA plasma (range 81-101%, n=5) and 84% in heparin plasma (range 80-89%, n=5), with dilution linearity characterised at 1:2, 1:4, 1:8 and 1:16 across these matrices.
