Mouse Actin Related Protein 2/3 Complex Subunit 4 ELISA Kit

Manufactured for Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of mouse Actin Related Protein 2/3 Complex Subunit 4 (ARPC4) in tissue homogenates or other biological fluids. The kit employs the sandwich enzyme immunoassay technique: an antibody specific for ARPC4 has been pre-coated onto a 96-well microplate, standards and test samples are added, and captured ARPC4 is detected using a Biotinylated Anti-ARPC4 Antibody followed by an HRP-Avidin conjugate. The enzymatic reaction is visualised with TMB substrate, producing a blue-coloured product that changes to yellow after addition of acidic stop solution, with absorbance read at 450 nm in a colorimetric microplate reader.

The assay has a sensitivity of 0.043 ng/ml, an assay range of 0.156–10 ng/ml, and a total assay time of 4 hours 30 minutes. Recovery testing (n=5 per matrix) gave averages of 91% for serum (range 80–102%), 90% for EDTA plasma (range 81–100%), and 84% for heparin plasma (range 80–89%). Dilution linearity at 1:2, 1:4, 1:8 and 1:16 (n=5) yielded: serum 87–91%, 87–107%, 74–101% and 92–97%; EDTA plasma 90–105%, 84–101%, 90–101% and 79–108%; heparin plasma 84–95%, 92–105%, 82–105% and 89–91%. Kit components include a pre-coated 96-well microplate, lyophilised standard, detection reagents, wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers, stored at +4°C or −20°C as instructed.
