Monkey Tumor Necrosis Factor Alpha ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of monkey Tumor Necrosis Factor Alpha (TNF alpha) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids.

The assay uses a 96-well microplate pre-coated with anti-TNF alpha antibody. Standards and samples are added to the wells, followed by a biotinylated anti-TNF alpha detection antibody and an HRP-avidin conjugate. A TMB substrate develops a colour change from blue to yellow on addition of the acidic stop solution; absorbance is then read at 450 nm in a colorimetric microplate reader and the TNF alpha concentration is calculated from a standard curve. The quantitative colourimetric assay is completed in approximately 4 hours 30 minutes, with a measuring range of 15.6-1000 pg/ml and a sensitivity of 6.9 pg/ml.

Validated spike-and-recovery across five serum samples averaged 91% (range 80-102%), with EDTA plasma averaging 90% (range 81-100%) and heparin plasma averaging 84% (range 80-89%). Parallelism has been demonstrated across 1:2 to 1:16 serial dilutions, with serum ranging 74-107%, EDTA plasma 79-108% and heparin plasma 82-105%. Kit components include a lyophilised TNF alpha standard, sample dilution buffer, concentrated wash buffer, two detection solutions, TMB substrate, stop solution and adhesive plate sealers, supplied for storage at +4°C or -20°C as instructed.
