Description

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of monkey TNF alpha in serum, plasma, tissue homogenates, and other biological fluids. The assay is specific for TNF alpha, with no reported cross-reactivity with other analogs or variants, and operates across a working range of 31.25–2000 pg/mL with a sensitivity below 18.75 pg/mL.

The procedure is carried out on a pre-coated 96-well microtiter plate arranged as 12 x 8 well strips and takes approximately four hours to complete. Detection is colorimetric, with absorbance read at 450 nm using a standard microplate reader. Standards and samples are incubated in the wells so that TNF alpha binds to the immobilised capture antibody; a biotinylated detection antibody is then added, followed by an HRP-Streptavidin conjugate and TMB substrate, producing a blue colour that turns yellow upon addition of the acidic stop solution. The intensity of the yellow product is directly proportional to the concentration of TNF alpha captured in each well.

Serum and plasma samples may affect test results and should be diluted at least 1:2 with Sample Dilution Buffer prior to testing; validated dilutions include healthy serum at 1:2 and undiluted monkey peripheral blood cell culture supernatants stimulated with 1 µg/mL LPS for 12 hours. Reported recovery averages are 95% for serum, 93% for EDTA plasma, and 90% for heparin plasma. The kit is shipped at +4 °C, with components stored at +4 °C or −20 °C as instructed; freeze/thaw cycles should be avoided and the kit should not be used past its expiration date.
