Human X-Ray Repair Cross Complementing 6 ELISA Kit

A sandwich ELISA from Antibodies.com for the in vitro quantitative determination of human Ku70 (X-Ray Repair Cross Complementing 6) in tissue homogenates, cell lysates, or other biological fluids. An antibody specific for the target is pre-coated onto a 96-well microplate. Standards and samples are added to the wells, where the target is captured by the immobilised antibody. After incubation and washing, a biotinylated anti-target antibody binds the captured target, followed by addition of an HRP-avidin conjugate. Following further incubation and washing, TMB substrate is added; the HRP-catalysed reaction produces a blue product that turns yellow upon addition of acidic stop solution. Absorbance is read at 450 nm in a colorimetric microplate reader, with the yellow intensity proportional to the amount of target captured, and concentration is determined against a standard curve.

The reported assay range is 0.312–20 ng/ml with a sensitivity of 0.112 ng/ml, and a typical total assay time of 4 h 30 min. Recovery has been demonstrated in serum (80%–102%, average 91%), EDTA plasma (81%–100%, average 90%) and heparin plasma (80%–89%, average 84%) at the dilutions evaluated. Kit components should be stored at +4°C or −20°C as instructed and not used past the stated expiration date.
