This sandwich Enzyme-Linked Immunosorbent Assay from Antibodies.com enables the in vitro quantitative determination of human X-Ray Repair Cross Complementing 5 (Ku80) in tissue homogenates or other biological fluids. A capture antibody specific for the target is pre-coated onto the microplate wells; standards and test samples are added, and the bound target is detected using a biotinylated anti-Ku80 antibody followed by an HRP-avidin conjugate. TMB substrate produces a blue colour that turns yellow upon addition of the stop solution, and the absorbance read at 450 nm is used to calculate target concentration against the standard curve.

The quantitative assay has a sensitivity of <0.057 ng/ml and a measurable range of 0.156–10 ng/ml, with a total run time of 4 hours 30 minutes. Reported average recovery from undiluted samples is 91% for serum (range 80–102%, n=5), 90% for EDTA plasma (range 81–100%, n=5) and 84% for heparin plasma (range 80–89%, n=5). Dilution linearity has been assessed at 1:2, 1:4, 1:8 and 1:16 across each matrix. Detection is colourimetric and components are stored at +4 °C or −20 °C as indicated; the kit should not be used past the printed expiration date.
