Human X-Linked Inhibitor Of Apoptosis Protein ELISA Kit

This sandwich ELISA from Antibodies.com is designed for the in vitro quantitative determination of human XIAP in tissue homogenates, cell lysates or other biological fluids. The assay uses the sandwich enzyme immunoassay technique: an antibody specific for XIAP is pre-coated onto the microplate, standards and samples are added, and after washing the wells are incubated with a biotinylated anti-XIAP antibody followed by an HRP-avidin conjugate. TMB substrate is then added; catalysis produces a blue product that turns yellow on addition of stop solution. The intensity of yellow is proportional to the XIAP captured, and concentration is calculated from the O.D. absorbance read at 450 nm against a standard curve.

Reported sensitivity is <0.103 ng/ml with an assay range of 0.312–20 ng/ml. The colourimetric, quantitative assay is run on a pre-coated microplate (12 x 8 well strips) and takes 4 hours 30 minutes. Kit components are stored at +4°C or −20°C as instructed, and the kit should not be used past its expiration date.

Average recovery from spiked samples was 91% in serum (range 80–102%, n=5), 90% in EDTA plasma (range 81–100%, n=5) and 84% in heparin plasma (range 80–89%, n=5).
