Human WNT Inhibitory Factor 1 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com, designed for the in vitro quantitative determination of human WIF1 (WNT Inhibitory Factor 1) in serum, plasma, tissue homogenates or other biological fluids. The format uses a 96-well microtiter plate pre-coated with an antibody specific for WIF1. Standards and samples are added to the wells, where WIF1 binds to the immobilised capture antibody. After washing, a biotinylated anti-WIF1 detection antibody is added, followed by HRP-avidin conjugate and TMB substrate. The enzymatic reaction produces a blue product that converts to yellow upon addition of the acidic stop solution; absorbance is read at 450 nm and the WIF1 concentration is calculated from the standard curve.

The assay is read on a colourimetric microplate reader and has a total run time of 4 hours 30 minutes. Reported sensitivity is <6.9 pg/ml and the standard curve range is 15.6-1000 pg/ml. Spike-and-recovery data are provided for serum (n=5, range 80%-102%, average 91%), EDTA plasma (n=5, range 81%-100%, average 90%) and heparin plasma (n=5, range 80%-89%, average 84%). Dilution linearity across 1:2, 1:4, 1:8 and 1:16 dilutions is reported for serum, EDTA plasma and heparin plasma (n=5 each). Kit components are stored at +4°C or -20°C as instructed in the protocol, and the product should not be used past its expiration date.
