Human Wiskott Aldrich Syndrome Protein Family, Member 2 ELISA Kit

This Antibodies.com sandwich ELISA kit is designed for the in vitro quantitative determination of human WASF2/WAVE2 (Wiskott Aldrich Syndrome Protein Family, Member 2) in tissue homogenates, cell lysates, or other biological fluids. The assay uses the sandwich enzyme immunoassay technique on a pre-coated 96-well microplate arranged as 12 x 8 well strips. Standards and samples are added to wells pre-coated with an antibody specific for WASF2/WAVE2, followed by incubation with a Biotinylated Anti-WASF2/WAVE2 Antibody and an HRP-Avidin conjugate. TMB substrate produces a blue colour that changes to yellow after addition of acidic stop solution. Absorbance is read at 450 nm in a colorimetric microplate reader, and the WASF2/WAVE2 concentration is calculated by reference to the standard curve.

Reported sensitivity is <0.106 ng/ml with an assay range of 0.312–20 ng/ml, and the total assay time is 4 h 30 min. The kit contains a pre-coated 96-well microplate, lyophilized standard, Detection Solutions A and B, 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution, and plate sealers, with components stored at +4 °C or –20 °C as instructed.

Spike-and-recovery in serum (n=5) averaged 91% (range 80%–102%); in EDTA plasma (n=5) it averaged 90% (range 81%–100%); and in heparin plasma (n=5) it averaged 84% (range 80%–89%). Dilution linearity across 1:2, 1:4, 1:8 and 1:16 dilutions yielded 87%–91%, 87%–107%, 74%–101% and 92%–97% in serum (n=5); 90%–105%, 84%–101%, 90%–101% and 79%–108% in EDTA plasma (n=5); and 84%–95%, 92%–105%, 82%–105% and 89%–91% in heparin plasma (n=5).
