Human Wingless Type MMTV Integration Site Family, Member 9B ELISA Kit

Antibodies.com provides a sandwich ELISA kit for the in vitro quantitative determination of human Wnt9b (Wingless Type MMTV Integration Site Family, Member 9B) in tissue homogenates, cell lysates, and other biological fluids. The assay is reactive with human samples and uses a pre-coated microplate format with 12 x 8 well strips, read colourmetrically at 450 nm.

The procedure follows a sandwich immunoassay technique. An antibody specific for Wnt9b is pre-coated onto the plate; standards and samples are added, and bound Wnt9b is detected using a biotinylated anti-Wnt9b antibody followed by HRP-avidin conjugate. TMB substrate produces a blue product that turns yellow upon addition of stop solution, with absorbance proportional to Wnt9b concentration. The kit has a sensitivity of <0.064 ng/ml and a working range of 0.156–10 ng/ml, with a total assay time of 4h 30m. Kit components are stored at +4°C or -20°C as indicated, and the kit should not be used past its expiration date.

Validation data report mean recoveries of 93% in serum (range 84–102%, n=5), 95% in EDTA plasma (range 90–101%, n=5), and 95% in heparin plasma (range 92–98%, n=5). Linearity across 1:2 to 1:16 dilutions was demonstrated for serum (range 75–104%), EDTA plasma (range 81–108%), and heparin plasma (range 79–105%).
