From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (ELISA) is designed for the in vitro quantitative determination of human Wnt7a (Wingless Type MMTV Integration Site Family, Member 7A) in tissue homogenates, cell lysates, and other biological fluids. The assay uses a pre-coated 96-well microplate with an antibody specific for Wnt7a immobilised on the wells. Standards and samples are added, followed by a biotinylated anti-Wnt7a detection antibody and an HRP-Avidin conjugate. TMB substrate visualises the reaction as a blue product that turns yellow upon addition of stop solution, and absorbance is read at 450 nm using a colorimetric microplate reader.

The quantitative assay has a reported sensitivity of <11.6 pg/ml and a standard range of 31.2–2000 pg/ml, with a total run time of 4 hours 30 minutes. Reported validation data include serum (range 83%–100%, average 91%, n=5), EDTA plasma (range 91%–100%, average 95%, n=5), and heparin plasma (range 91%–101%, average 96%, n=5), with linearity data provided across 1:2, 1:4, 1:8, and 1:16 dilutions.
