Human Wingless Type MMTV Integration Site Family, Member 5A ELISA Kit

Manufactured for Antibodies.com, this Sandwich ELISA kit enables the in vitro quantitative determination of human Wnt5a (Wingless Type MMTV Integration Site Family, Member 5A) in tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay is configured for use with human samples and is read on a colorimetric microplate reader.

The kit applies the sandwich enzyme immunoassay technique using a microplate pre-coated with an antibody specific for Wnt5a. Standards and test samples are added to the wells and bound by the immobilised antibody, followed by incubation with a Biotinylated Anti-Wnt5a Antibody and then an HRP-Avidin conjugate. TMB substrate is used to visualise the enzymatic reaction, producing a blue colour that changes to yellow on addition of acidic stop solution. Absorbance is read at 450 nm and Wnt5a concentration is calculated against the standard curve. Total assay time is 4 hours 30 minutes.

The standard curve covers 0.156–10 ng/ml with a sensitivity of 0.072 ng/ml. Reported average recovery is 91% for serum (range 83–100%, n=5), 94% for EDTA plasma (range 90–99%, n=5) and 96% for heparin plasma (range 91–101%, n=5). Components are stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
