Antibodies.com supplies this sandwich Enzyme-Linked Immunosorbent Assay (ELISA) kit for the in vitro quantitative determination of human Wnt2/IRP (Wingless Type MMTV Integration Site Family, Member 2) in tissue homogenates, cell lysates or other biological fluids. The assay employs a pre-coated microplate (12 x 8 well strips) to which standards and samples are added; captured Wnt2/IRP is detected with a Biotinylated Anti-Wnt2/IRP Antibody followed by an HRP-Avidin conjugate. TMB substrate produces a blue-coloured product that changes to yellow on addition of acidic stop solution, and the O.D. absorbance at 450 nm is read on a colourmetric microplate reader.

The quantitative sandwich format has an assay range of 0.156-10 ng/ml and a sensitivity of <0.062 ng/ml, with a total assay time of 4h 30m. Recovery has been characterised across multiple sample matrices: serum (n=5) showed 80-102% recovery with an average of 91%; EDTA plasma (n=5) showed 81-100% with an average of 90%; heparin plasma (n=5) showed 80-89% with an average of 84%. Dilution linearity was assessed at 1:2, 1:4, 1:8 and 1:16, with serum (n=5) at 87-91%, 87-107%, 74-101% and 92-97%; EDTA plasma (n=5) at 90-105%, 84-101%, 90-101% and 79-108%; and heparin plasma (n=5) at 84-95%, 92-105%, 82-105% and 89-91%.
