From Antibodies.com, this Human Wnt11 ELISA Kit is a sandwich enzyme-linked immunosorbent assay (sELISA) intended for the in vitro quantitative determination of human Wingless Type MMTV Integration Site Family, Member 11 (Wnt11) in tissue homogenates, cell lysates, cell culture supernatants, or other biological fluids. The product is for research use only and is not intended for diagnostic or therapeutic applications.

The assay uses a pre-coated 96-well microplate arranged as 12 x 8 well strips. An antibody specific for Wnt11 captures the target from standards and samples, followed by incubation with a Biotinylated Anti-Wnt11 Antibody and an HRP-Avidin conjugate. A TMB substrate solution produces a colour change that is read at 450 nm on a colorimetric microplate reader, with the Wnt11 concentration calculated against the standard curve. Total assay time is 4h 30m. Sensitivity is <0.058 ng/ml and the working range spans 0.156–10 ng/ml.

Kit components include a pre-coated microplate, lyophilized standard, Detection Solution A, Detection Solution B, 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution, and plate sealers, stored at +4°C or –20°C as instructed. Average spike recovery was reported as 91% in serum (n=5, 83–100%), 96% in EDTA plasma (n=5, 90–103%), and 95% in heparin plasma (n=5, 89–102%). Dilution linearity across 1:2, 1:4, 1:8 and 1:16 dilutions gave recovery ranges of 83–110%, 79–100%, 90–98% and 82–108% for serum; 90–98%, 90–105%, 73–109% and 87–91% for EDTA plasma; and 88–103%, 77–105%, 95–98% and 87–110% for heparin plasma.
