Manufactured by Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of human Wilms Tumor Protein in tissue homogenates, cell lysates, or other biological fluids. The product is intended for research use only and is not for diagnostic or therapeutic use.

The assay employs the sandwich enzyme immunoassay technique using a pre-coated microplate. An antibody specific for Wilms Tumor Protein is pre-coated onto the plate; standards and samples are added and the target is captured. Following incubation and washing, a Biotinylated Anti-Wilms Tumor Protein Antibody and HRP-Avidin conjugate are added sequentially. TMB substrate solution is then used to visualise the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of Wilms Tumor Protein captured, measured by O.D. absorbance at 450nm in a microplate reader and calculated against a standard curve. Performance specifications include an assay range of 0.156-10 ng/ml and a sensitivity of <0.059 ng/ml, with a total assay time of 4h 30m.

Sample validation data report average recovery of 91% for serum (range 80-102%), 90% for EDTA plasma (range 81-100%), and 84% for heparin plasma (range 80-89%), with linearity confirmed across dilutions of 1:2 to 1:16. Kit components include a pre-coated microplate, lyophilized standard, Detection Solutions A and B, 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution, and plate sealers, to be stored at +4°C or -20°C as instructed.
