Human Von Willebrand Factor A Domain Containing Protein 2 ELISA Kit

This sandwich Enzyme-Linked Immunosorbent Assay (sELISA), distributed by Antibodies.com, is designed for the in vitro quantitative determination of human AMACO (Von Willebrand Factor A Domain Containing Protein 2) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids.

The assay uses the sandwich enzyme immunoassay technique. An antibody specific for AMACO is immobilised on a pre-coated microplate. Standards and test samples are added and the target analyte in each sample binds to the immobilised antibody. After incubation and washing, a Biotinylated Anti-AMACO Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate is then added and the HRP enzymatic reaction produces a blue product that changes to yellow on addition of the acidic stop solution. The intensity of yellow colour is proportional to the amount of AMACO captured, and absorbance is read at 450 nm in a microplate reader with concentration calculated by reference to a standard curve.

Key performance data from the manufacturer source include a detection range of 1.56–100 ng/ml and a sensitivity of <0.6 ng/ml. The assay is completed in 4 hours 30 minutes and is intended for use on a colorimetric microplate reader. Reported spike-and-recovery values across serum, EDTA plasma and heparin plasma range from 80% to 102% with averages between 84% and 91%. Reported values across 1:2 to 1:16 dilutions of the same sample types range from 74% to 108%. Kit components are stored at +4°C or -20°C as instructed, and the kit must not be used past its expiration date.
