Human Vitronectin ELISA Kit

This sandwich ELISA from Antibodies.com is intended for the in vitro quantitative determination of human Vitronectin (also referred to as S-Protein) in serum, plasma or other biological fluids. The kit employs the sandwich enzyme immunoassay technique, using an antibody specific for Vitronectin/S-Protein pre-coated onto a 96-well microtiter plate. Standards and test samples are added to the wells, captured analyte is detected with a Biotinylated Anti-Vitronectin/S-Protein Antibody followed by an HRP-Avidin conjugate, and the reaction is visualised with a TMB substrate solution that turns yellow after addition of stop solution. Absorbance is read at 450 nm in a colorimetric microplate reader.

The assay has a working range of 3.12–200 ng/ml and a sensitivity of <1.44 ng/ml, with a total assay time of 4 hours 30 minutes. The platform is a pre-coated microplate arranged as 12 x 8-well strips, and kit components are stored at +4°C or –20°C as instructed.

Spike-and-recovery performance was assessed across serum, EDTA plasma and heparin plasma (n=5 each), with average recoveries of 91% (serum, range 80–102%), 90% (EDTA plasma, range 81–100%) and 84% (heparin plasma, range 80–89%). Linearity was demonstrated at dilutions of 1:2, 1:4, 1:8 and 1:16 in all three matrices.
