From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of human Viperin in tissue homogenates, cell lysates, or other biological fluids. An antibody specific for Viperin is pre-coated onto a 96-well microplate; standards and samples are added, followed by a Biotinylated Anti-Viperin Antibody and HRP-Avidin conjugate. TMB substrate produces a blue product that turns yellow after addition of acidic stop solution, with absorbance at 450 nm proportional to Viperin concentration, calculated from a standard curve.

The colorimetric assay has a sensitivity of <0.062 ng/ml and an assay range of 0.156-10 ng/ml, with a total run time of 4 hours 30 minutes on a pre-coated 12 x 8 well strip microplate read on a colorimetric microplate reader. Kit components include the pre-coated plate, lyophilized standard (2 vials), detection solutions A and B, 30X wash buffer, sample dilution buffer, TMB substrate, stop solution, and adhesive plate sealers, stored at +4°C or -20°C as instructed. Recovery was assessed in spiked samples with average values of 93% in serum (n=5, range 86-100%), 94% in EDTA plasma (87-102%), and 94% in heparin plasma (88-100%). Dilution linearity was demonstrated at 1:2, 1:4, 1:8, and 1:16 across the same sample types.
