Human Vinculin ELISA Kit

This sandwich ELISA kit from Antibodies.com is designed for the in vitro quantitative determination of human Vinculin in serum, plasma, tissue homogenates and other biological fluids. The assay uses a pre-coated 96-well microplate configured as 12 x 8 well strips. Standards and samples are added to the wells where Vinculin is captured by the immobilized antibody, followed by incubation with a biotinylated anti-Vinculin detection antibody. An HRP-Avidin conjugate is then added, and the enzymatic reaction is visualised with TMB substrate, producing a yellow product whose intensity is read at 450 nm on a colorimetric microplate reader.

The quantitative assay has a reported sensitivity of <32 pg/ml and a measuring range of 78.1-5000 pg/ml, with a total incubation time of approximately 4 hours 30 minutes. Reported spike-and-recovery values for undiluted samples averaged 91% in serum (range 80-102%), 90% in EDTA plasma (range 81-100%) and 84% in heparin plasma (range 80-89%). Linearity across 1:2 to 1:16 dilutions has been demonstrated, with recoveries generally between 74% and 108% depending on sample type.

The kit is supplied with the reagents required to perform the sandwich ELISA workflow, including the pre-coated microplate, lyophilized standard, biotinylated detection reagent, wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers. Components should be stored at +4°C or -20°C as indicated in the protocol, and the kit should not be used past its expiration date.
