Human Vasoactive Intestinal Peptide ELISA Kit

The kit employs a competitive enzyme immunoassay technique on a 96-well microtiter plate pre-coated with VIP antigen. During incubation, VIP present in samples or standards competes with the immobilized VIP for binding sites on the Biotinylated Anti-VIP Antibody. After washing to remove unbound antibody, an HRP-Avidin conjugate is added, and TMB substrate solution is used to visualize the reaction. Following addition of acidic stop solution, the colour change is read by absorbance at 450 nm in a colorimetric microplate reader; the density of yellow is inversely proportional to the VIP concentration, which is calculated against the standard curve. The product range is 7.81-500 pg/ml, with a sensitivity of 2.34 pg/ml, and the total assay time is approximately 2 hours 30 minutes.

Validated sample recoveries average 91% for serum (range 80-102%, n=5), 90% for EDTA plasma (range 81-100%, n=5) and 84% for heparin plasma (range 80-89%, n=5). Dilution linearity has been demonstrated across 1:2, 1:4, 1:8 and 1:16 dilutions for serum, EDTA plasma and heparin plasma (n=5 each). Kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
