The assay uses the sandwich enzyme immunoassay technique, with an antibody specific for VEGF Receptor 1 pre-coated onto a 96-well microplate. Standards and test samples are added to the wells, and captured VEGF Receptor 1 is detected using a biotinylated anti-VEGF Receptor 1 antibody followed by an HRP-avidin conjugate.

Quantitative measurement is achieved using a TMB substrate, producing a yellow product whose intensity is read at an absorbance of 450 nm in a colorimetric microplate reader. The reported assay range is 0.156–10 ng/ml with a sensitivity of 0.066 ng/ml. The platform is a pre-coated microplate of 12 × 8 well strips and the total assay time is approximately 4 hours 30 minutes.
