This sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is designed for the in vitro quantitative determination of human c-Myc, also referred to as V-Myc Myelocytomatosis Viral Oncogene Homolog, in tissue homogenates, cell lysates and other biological fluids. The kit uses the sandwich enzyme immunoassay technique on a 96-well microplate pre-coated with an antibody specific for c-Myc. Standards and test samples are added to the wells, the plate is washed, and a Biotinylated Anti-c-Myc Antibody is applied to bind captured c-Myc. HRP-Avidin conjugate is then added, followed by TMB substrate solution; the resulting blue-coloured product turns yellow after the addition of acidic stop solution. Absorbance is read at 450 nm in a microplate reader and the c-Myc concentration is calculated by reference to a standard curve. The detection method is colourmetric, the total assay time is 4h 30m, and kit components should be stored at +4°C or -20°C as instructed.

Reported sensitivity is <0.054 ng/ml with an assay range of 0.156–10 ng/ml. Validation recovery data are provided for serum, EDTA plasma and heparin plasma. Mean recoveries across sample matrices are 91% for serum (range 80–102%, n=5), 90% for EDTA plasma (range 81–100%, n=5) and 84% for heparin plasma (range 80–89%, n=5). Dilution linearity has been assessed at 1:2, 1:4, 1:8 and 1:16 dilutions, with overall recovery ranges of 74–107% for serum, 79–108% for EDTA plasma and 82–105% for heparin plasma depending on dilution factor.
