A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com, designed for the in vitro quantitative determination of human UACA/Nucling (Uveal Autoantigen With Coiled Coil Domains And Ankyrin Repeats) in tissue homogenates or other biological fluids.
The assay employs the sandwich enzyme immunoassay technique using a pre-coated 96-well microplate. An antibody specific for UACA/Nucling is immobilised on the plate; standards and samples are added and the target binds to the captured antibody. After washing, a Biotinylated Anti-UACA/Nucling Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate produces a blue-coloured product that turns yellow upon addition of acidic stop solution. Absorbance is read at 450 nm in a microplate reader and the target concentration is calculated from the standard curve.
Reported sensitivity is <0.132 ng/ml and the assay range is 0.312–20 ng/ml, with a total assay time of approximately 4 hours 30 minutes. The plate is read on a colorimetric microplate reader and components should be stored at +4°C or −20°C as indicated. Validation data for serum, EDTA plasma, and heparin plasma samples at dilution factors of 1:2, 1:4, 1:8, and 1:16 are provided by the manufacturer.
