Human Uromodulin ELISA Kit

Manufactured for Antibodies.com, this Human Uromodulin ELISA Kit is a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) designed for the in vitro quantitative determination of human UMOD in serum, plasma, urine or other biological fluids. The assay employs the sandwich enzyme immunoassay technique, with an antibody specific for UMOD pre-coated onto a microtiter plate. Standards and test samples are added to the wells, where UMOD is captured by the immobilised antibody. After washing, a Biotinylated Anti-UMOD Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate is then used to visualise the HRP enzymatic reaction, producing a blue colour that changes to yellow upon addition of stop solution. The absorbance is read at 450 nm in a microplate reader and the UMOD concentration calculated by reference to a standard curve.

Reported sensitivity is <1.25 ng/ml with an assay range of 3.12–200 ng/ml, read on a colorimetric microplate reader. The pre-coated microplate is supplied as 12 x 8 well strips and the total assay time is 4 hours 30 minutes. Kit components include pre-coated microplate strips, lyophilised standard, Detection Solutions A and B, Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution and plate sealers, stored at +4°C or −20°C as instructed. The kit should not be used past its expiration date.

Spike-and-recovery testing reported average recoveries of 91% in serum (n=5, range 80–102%), 90% in EDTA plasma (n=5, range 81–100%) and 84% in heparin plasma (n=5, range 80–89%). Dilution linearity was demonstrated at 1:2, 1:4, 1:8 and 1:16 across serum, EDTA plasma and heparin plasma samples (n=5 each).
