Human Uncoupling Protein 2, Mitochondrial ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of human Uncoupling Protein 2 (UCP2) in tissue homogenates or other biological fluids. The plate is pre-coated with an antibody specific for UCP2. Standards and test samples are added to the wells, and the captured UCP2 is bound by a Biotinylated Anti-UCP2 Antibody added after washing. An HRP-Avidin conjugate is then introduced and TMB substrate is added to visualise the enzymatic reaction. The colour intensity is proportional to the amount of UCP2 captured and is read at 450 nm in a colorimetric microplate reader, with the concentration calculated by reference to a standard curve.

Reported sensitivity is <0.051 ng/ml and the assay range is 0.156–10 ng/ml. The complete assay takes 4 hours 30 minutes. Reported recovery in spiked samples is: Serum (n=5) 80%–102% averaging 91%; EDTA Plasma (n=5) 81%–100% averaging 90%; Heparin Plasma (n=5) 80%–89% averaging 84%. Dilution linearity across 1:2, 1:4, 1:8 and 1:16 is reported as: Serum 87–91%, 87–107%, 74–101%, 92–97%; EDTA Plasma 90–105%, 84–101%, 90–101%, 79–108%; Heparin Plasma 84–95%, 92–105%, 82–105%, 89–91%.

The kit contains a pre-coated 96-well microplate, lyophilized standard, detection solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution and adhesive plate sealers. Components are stored between +4°C and −20°C as instructed, and the kit should not be used past its expiration date.
