Human Ubiquitin Specific Peptidase 8 ELISA Kit

From Antibodies.com, this sandwich ELISA kit is intended for the in vitro quantitative determination of human UBPY/USP8 in tissue homogenates, cell lysates or other biological fluids. The assay uses a pre-coated 96-well microplate with an immobilised antibody specific for UBPY/USP8. Standards and samples are added to the wells, followed by a biotinylated anti-UBPY/USP8 antibody and an HRP-avidin conjugate. After incubation with TMB substrate, the reaction produces a blue colour that turns yellow upon addition of stop solution; absorbance is read at 450nm on a colorimetric microplate reader and the UBPY/USP8 concentration is calculated from the standard curve.

The reported assay range is 0.312-20 ng/ml with a sensitivity of 0.101 ng/ml, and total assay time is 4h 30m. Recovery testing in serum (n=5) returned 80%-102% (average 91%); EDTA plasma (n=5) 81%-100% (average 90%); and heparin plasma (n=5) 80%-89% (average 84%). Dilution linearity across 1:2, 1:4, 1:8 and 1:16 dilutions was 87-91%, 87-107%, 74-101% and 92-97% respectively for serum; 90-105%, 84-101%, 90-101% and 79-108% for EDTA plasma; and 84-95%, 92-105%, 82-105% and 89-91% for heparin plasma. Kit components should be stored at +4°C or -20°C as instructed and the product should not be used past its expiration date.
