Human Ubiquitin Conjugating Enzyme E2I ELISA Kit

This sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is designed for the in vitro quantitative determination of human UBE2I/UBC9 (Ubiquitin Conjugating Enzyme E2I) in tissue homogenates, cell lysates or other biological fluids. The assay employs the sandwich enzyme immunoassay technique, using an antibody specific for UBE2I/UBC9 that has been pre-coated onto a microplate. Standards and test samples are added to the wells and UBE2I/UBC9 present in each sample is captured by the immobilised antibody. After washing, a Biotinylated Anti-UBE2I/UBC9 Antibody is added, followed by an HRP-Avidin conjugate. A TMB substrate solution is then used to visualise the reaction, producing a blue colour that changes to yellow upon addition of acidic stop solution. Absorbance is read at 450 nm in a colorimetric microplate reader and the concentration is calculated against the standard curve [S1].

The kit has a stated sensitivity of <0.053 ng/ml and an assay range of 0.156-10 ng/ml, with a total assay time of 4h 30m [S1]. Reactivity is human [C0, S1]. Sample recovery was assessed in serum (average 91%, range 83-100%, n=5), EDTA plasma (average 96%, range 90-103%, n=5) and heparin plasma (average 94%, range 90-99%, n=5) [S1]. Dilution linearity was reported at 1:2, 1:4, 1:8 and 1:16 across serum, EDTA plasma and heparin plasma [S1].

Components supplied include a pre-coated microplate arranged as 12 x 8 well strips, lyophilised standard, Detection Solutions A and B, Wash Buffer (30X), Sample Dilution Buffer, TMB Substrate, Stop Solution and plate sealers.
