The assay employs the sandwich enzyme immunoassay technique on a pre-coated 96-well microplate arranged as 12 x 8 well strips, using a biotinylated anti-TIE1 detection antibody and an HRP-avidin conjugate with TMB substrate. Signal is read at 450 nm on a colorimetric microplate reader, with a total assay time of 4 hours 30 minutes.

Reported sensitivity is <0.62 ng/ml with a product range of 1.56–100 ng/ml. Spike-and-recovery testing (n=5) gave average recoveries of 92% in serum (range 84–100%), 91% in EDTA plasma (83–99%) and 93% in heparin plasma (85–101%). Dilution linearity was demonstrated across 1:2, 1:4, 1:8 and 1:16 sample dilutions, with serum values of 85–110%, 77–102%, 90–96% and 84–108%; EDTA plasma values of 88–100%, 90–104%, 76–110% and 86–93%; and heparin plasma values of 89–102%, 75–106%, 94–97% and 88–110%.
