Human Tyrosine 3/Tryptophan 5 Monooxygenase Activation Protein Zeta ELISA Kit

A sandwich enzyme-linked immunosorbent assay (ELISA) kit from Antibodies.com designed for the in vitro quantitative determination of human 14-3-3 zeta (Tyrosine 3/Tryptophan 5 Monooxygenase Activation Protein Zeta) in serum, plasma, tissue homogenates, cell lysates or other biological fluids. The assay employs the sandwich immunoassay technique, using an antibody specific for 14-3-3 zeta pre-coated onto a 96-well microplate. Standards and test samples are added to the wells and any 14-3-3 zeta present is bound by the immobilised antibody. After washing, a biotinylated anti-14-3-3 zeta antibody is added, followed by an HRP-avidin conjugate. TMB substrate is then used to visualise the HRP enzymatic reaction, producing a blue product that turns yellow after the addition of acidic stop solution. The density of yellow is proportional to the amount of 14-3-3 zeta captured, and concentration is calculated by reading the O.D. absorbance at 450 nm in a microplate reader against a standard curve.

The kit has a product range of 0.156–10 ng/ml and a reported sensitivity of <0.053 ng/ml, with a total assay time of 4 h 30 min. Recovery has been demonstrated in serum (range 80%–102%, average 91%, n=5), EDTA plasma (range 81%–100%, average 90%, n=5) and heparin plasma (range 80%–89%, average 84%, n=5). Kit components are stored at +4 °C or -20 °C as instructed, and the product should not be used past its expiration date. The ready-to-use kit is suitable for serum, plasma, cell lysates and tissue homogenates.
