Human Twist Transcription Factor ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) supplied by Antibodies.com, designed for the in vitro quantitative determination of human Twist Transcription Factor in tissue homogenates, cell lysates and other biological fluids. The assay uses a pre-coated 96-well microplate arranged in a 12 x 8 well strip format. Standards and samples are added to the wells, where Twist is captured by an immobilised specific antibody. After washing, a Biotinylated Anti-Twist Antibody is added and binds the captured target, followed by HRP-Avidin conjugate. TMB substrate is then added to visualise the HRP enzymatic reaction, producing a blue-coloured product that turns yellow on addition of the acidic stop solution; absorbance is read at 450 nm in a colorimetric microplate reader and the concentration of Twist is calculated against the standard curve.

The quantitative sandwich assay has a reported sensitivity of <0.055 ng/ml and an assay range of 0.156-10 ng/ml, with a total assay time of 4h 30m. Validation data for serum, EDTA plasma and heparin plasma indicate recovery ranges of 80%-102% (average 91%, n=5), 81%-100% (average 90%, n=5) and 80%-89% (average 84%, n=5) respectively. Linearity has been reported across 1:2, 1:4, 1:8 and 1:16 dilutions for each sample type. Kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
