Human Twinfilin 1 ELISA Kit

A sandwich ELISA from Antibodies.com is designed for the in vitro quantitative determination of human TWF1/Twinfilin-1 in tissue homogenates or other biological fluids. The assay uses a pre-coated 96-well microplate (12 x 8 well strips) bearing an antibody specific for TWF1/Twinfilin-1. Standards and test samples are added to the wells, captured TWF1/Twinfilin-1 is bound by a biotinylated anti-TWF1/Twinfilin-1 antibody, and an HRP-Avidin conjugate is added. TMB substrate is then used to visualise the HRP enzymatic reaction, producing a blue product that changes to yellow after addition of the acidic stop solution. Absorbance is read at 450 nm in a colorimetric microplate reader and the TWF1/Twinfilin-1 concentration is calculated from the standard curve.

The kit has a reported sensitivity of <0.054 ng/ml and an assay range of 0.156–10 ng/ml. The total assay time is 4 h 30 min. Kit components should be stored at +4°C or -20°C as instructed and not used past the expiration date.

Spike-and-recovery was evaluated in serum (n=5, range 80%–102%, average 91%), EDTA plasma (n=5, range 81%–100%, average 90%) and heparin plasma (n=5, range 80%–89%, average 84%). Linearity-of-dilution was assessed at 1:2, 1:4, 1:8 and 1:16 dilutions, with recoveries of 87–91%, 87–107%, 74–101% and 92–97% for serum; 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma; and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma (all n=5).
