From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of human TNFRSF1B in serum, plasma, tissue homogenates or other biological fluids. The kit employs the sandwich enzyme immunoassay technique using a 96-well microtiter plate pre-coated with an antibody specific for TNFRSF1B. Standards and test samples are added to the wells; after incubation and washing, a Biotinylated Anti-TNFRSF1B Antibody and an HRP-Avidin conjugate are applied. A TMB substrate solution is then used to visualise the enzymatic reaction, producing a blue product that turns yellow on addition of acidic stop solution. The intensity of the yellow colour is proportional to the amount of TNFRSF1B captured, and the concentration is calculated by reading the O.D. absorbance at 450 nm in a microplate reader against a standard curve.

The assay has a reported range of 0.156-10 ng/ml and a sensitivity of <0.063 ng/ml, with a total assay time of 4 h 30 min. Average recovery values (n=5) are 91% for serum (range 83-100%), 94% for EDTA plasma (range 90-98%), and 95% for heparin plasma (range 89-102%). Dilution linearity across 1:2, 1:4, 1:8 and 1:16 dilutions (n=5) is reported as 83-110%, 79-100%, 90-98% and 82-108% for serum; 90-98%, 90-105%, 73-109% and 87-91% for EDTA plasma; and 88-103%, 77-105%, 95-98% and 87-110% for heparin plasma. Kit components are stored at +4°C or -20°C as instructed, and the kit is suitable for use with serum, plasma and tissue homogenates.
