The assay is reactive with human samples and is run on a pre-coated 96-well microplate arranged as 12 x 8 well strips, read on a colorimetric microplate reader at 450 nm.

An antibody specific for 4-1BBL is immobilised on the plate; standards and samples are added, followed by a biotinylated anti-4-1BBL detection antibody and an HRP-Avidin conjugate. TMB substrate produces a blue colour that turns yellow upon addition of the acidic stop solution, with the intensity of yellow proportional to the amount of 4-1BBL captured. The product range is 15.6-1000 pg/ml with a sensitivity of <5.35 pg/ml, and the total assay time is 4 hours 30 minutes.

Validated recovery data are reported for serum (80%-102%, n=5, average 91%), EDTA plasma (81%-101%, n=5, average 91%), and heparin plasma (80%-89%, n=5, average 84%).
