Human Tumor Necrosis Factor beta ELISA Kit

Manufactured by Antibodies.com, this sandwich ELISA kit is designed for the in vitro quantitative determination of human Tumor Necrosis Factor beta (TNF-β) in serum, plasma, and cell culture supernatant samples. The assay is reactive with human samples and has shown no cross-reactivity with various other cytokines super-family proteins. It is able to detect both natural and recombinant human TNF-β.

The kit employs a quantitative sandwich immunoassay format on a pre-coated microplate (12 x 8 well strips). A biotin-conjugated polyclonal antibody specific to TNF-β is used, together with Avidin conjugated to Horseradish Peroxidase (HRP) and a TMB substrate solution. The enzyme–substrate reaction is terminated with sulphuric acid and the colour change is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm using a colorimetric microplate reader. Total assay time is 3 hours. The kit is shipped and stored at 4°C.

Two calibration diluents are included: Calibrator Diluent I for serum and plasma testing, and Calibrator Diluent II for cell culture supernatant testing. The supplied standard is diluted 2-fold with the appropriate diluent to generate a standard curve of Optical Density versus TNF-β concentration (pg/mL). The reported sensitivity is < 11 pg/ml and the assay range is 11–8000 pg/ml.
