From Antibodies.com, this Sandwich ELISA kit is intended for the in vitro quantitative determination of human TNFAIP2 in serum, plasma, tissue homogenates or other biological fluids, with reactivity against Human samples. Detection is performed on a Colorimetric Microplate Reader.

The assay employs the sandwich enzyme immunoassay technique. A TNFAIP2-specific antibody is pre-coated onto a 96-well microplate arranged as 12 x 8 well strips. Standards and test samples are added; TNFAIP2 present is captured by the immobilized antibody. After washing, a Biotinylated Anti-TNFAIP2 Antibody is added, followed by an HRP-Avidin conjugate and TMB substrate. The enzymatic reaction produces a blue product that turns yellow upon addition of acidic stop solution; absorbance is read at 450 nm and the TNFAIP2 concentration is calculated from a standard curve. Total assay time is 4h 30m. The stated assay range is 0.312-20 ng/ml and sensitivity is <0.109 ng/ml.

Kit components are stored at +4°C or -20°C as instructed and the kit must not be used past its expiration date. Reported validation includes recovery across n=5 samples each of serum (range 80%-102%, average 91%), EDTA plasma (range 81%-101%, average 91%) and heparin plasma (range 80%-89%, average 84%). Dilution linearity at 1:2, 1:4, 1:8 and 1:16 is also reported across these same matrices.
