Human Tumor Necrosis Factor Alpha ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (ELISA) from Antibodies.com is designed for the in vitro quantitative determination of human Tumor Necrosis Factor Alpha (TNF alpha) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay uses a pre-coated microplate with an antibody specific for TNF alpha and operates on a colorimetric detection principle read at 450 nm using a standard microplate reader.

The quantitative standard curve covers a range of 15.6-1000 pg/ml, with a sensitivity of <6.62 pg/ml, and a total assay time of 4h 30m. Captured TNF alpha is detected using a Biotinylated Anti-TNF alpha Antibody followed by an HRP-Avidin conjugate and TMB substrate; the resulting yellow product intensity is proportional to TNF alpha concentration and is interpolated against the standard curve.

Validation in human serum and plasma shows the following recovery ranges: serum 80%-102% (average 91%), EDTA plasma 81%-100% (average 90%), and heparin plasma 80%-89% (average 84%). Linearity has been demonstrated across serial dilutions of 1:2, 1:4, 1:8 and 1:16 for serum, EDTA plasma and heparin plasma. Kit components should be stored at +4°C or -20°C as instructed and the product should not be used past its expiration date.
