Antibodies.com supplies a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) configured for the in vitro quantitative determination of human beta Tubulin in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids.

The assay employs the sandwich enzyme immunoassay technique. An antibody specific for beta Tubulin is pre-coated onto a microtiter plate; standards and test samples are added and the target analyte is captured by the immobilized antibody. After washing, a Biotinylated Anti-beta Tubulin Antibody is added, followed by HRP-Avidin conjugate. TMB substrate produces a blue colour that becomes yellow upon addition of acidic stop solution; the yellow intensity is proportional to beta Tubulin captured in each well. Concentration is calculated by reading O.D. absorbance at 450nm in a microplate reader against the standard curve.

Performance characteristics include a measuring range of 0.156–10 ng/ml and a sensitivity of 0.069 ng/ml. Validation recovery (n=5) showed: Serum 80–102% (average 91%), EDTA Plasma 81–100% (average 90%), Heparin Plasma 80–89% (average 84%). Dilution linearity was demonstrated at 1:2, 1:4, 1:8 and 1:16 across all three matrices. The assay is read on a Colorimetric Microplate Reader, runs for 4h 30m, and kit components are stored at +4°C or −20°C as instructed.
