Human Tryptophan Hydroxylase 2 ELISA Kit

Manufactured by Antibodies.com as part of their Assays and Kits range, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of human Tryptophan Hydroxylase 2 (TPH2) in serum, plasma, tissue homogenates or other biological fluids, with reactivity confirmed for human samples.

The assay uses a pre-coated 96-well microplate (12 x 8 well strips) with colourmetric detection read at 450 nm in a microplate reader and runs for approximately 4 hours 30 minutes. Standards and test samples are added to wells where TPH2 is captured by an immobilised antibody; binding is then detected using a Biotinylated Anti-TPH2 Antibody followed by an HRP-Avidin conjugate and TMB substrate, with the assay range quoted at 0.312-20 ng/ml and sensitivity of <0.127 ng/ml. Recovery data supplied by the manufacturer (n=5 per sample type) show serum 80%-102% (average 91%), EDTA plasma 81%-100% (average 90%), and heparin plasma 80%-89% (average 84%). Dilution linearity across 1:2, 1:4, 1:8 and 1:16 dilutions (n=5) is reported as: serum 87-91%, 87-107%, 74-101% and 92-97%; EDTA plasma 90-105%, 84-101%, 90-101% and 79-108%; heparin plasma 84-95%, 92-105%, 82-105% and 89-91%.

Storage requirements vary by component: the pre-coated 96 well microplate, lyophilised standard (2 vials), detection solution A (120μl), detection solution B (120µl) and sample dilution buffer (45ml) are stored at -20°C, while the 30X wash buffer (20ml), TMB substrate (9ml) and stop solution (6ml) are stored at +4°C. Five adhesive plate sealers are included, and kit components must not be used past their expiration date.
