Human Tryptase ELISA Kit

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of human Mast Cell Tryptase in serum, plasma, or other biological fluids. The procedure uses wells pre-coated with an antibody specific for Mast Cell Tryptase, into which standards and test samples are added. Following incubation and washing, a biotinylated anti-Mast Cell Tryptase antibody binds the captured antigen, after which an HRP-avidin conjugate and TMB substrate are added. The substrate reacts to produce a blue product that turns yellow on addition of acidic stop solution, with the intensity of yellow proportional to the tryptase concentration and quantified by reading absorbance at 450 nm on a colorimetric microplate reader.

Reported sensitivity is 0.125 ng/ml and the assay range is 0.312-20 ng/ml, with a complete assay time of approximately 4 hours 30 minutes. Spiking recovery (n=5) is 80%-102% (average 91%) in serum, 81%-100% (average 90%) in EDTA plasma, and 80%-89% (average 84%) in heparin plasma. Dilution linearity has been demonstrated for serum, EDTA plasma, and heparin plasma samples across serial dilutions.
